Modeling and simulation of droplet translocation and fission by electrowetting-on-dielectrics (EWOD)

摘要: This paper discusses methods of microfluidic droplet actuation by means of electrowetting-on-dielectrics (EWOD) and provides a technique for modeling and simulating a microfluidic device by using the computational fluid dynamics (CFD) program, Flow3D. Digital or droplet microfluidics implies the manipulation of droplets on a scale of nanoliters (10 L) to femtoliters (10 L), as opposed to continuous microfluidics that involve the control of continuous fluid within a channel. The two operations in focus here are droplet translocation (moving) and droplet fission (splitting), in which the pressures and velocities within the droplet are analyzed and compared to existing works, both theoretical and experimental. The variation in the pressure of the leading and trailing faces of a droplet indicates the variation in surface energy—an important parameter that explains how a droplet will move toward a region of higher electric potential. The higher voltage on one side of a droplet reduces surface energy, which leads to an induced pressure drop, thus resulting in fluid motion. Flow3D simulations are for both water and blood droplets at voltages between 50 V and 200 V, and the droplet size, surface properties (Teflon coated), and geometry of the system are kept constant for each operation. Some peculiarities of the simulation are brought to light, such as instabilities of the system to higher voltages and fluids with higher dielectric constants, as well as the creation of a tertiary droplet when the applied voltage causes a large enough force during fission. The force distribution across the droplet provides a general understanding of the electrowetting effect and more specifically allows for a comparison between the effects that different voltages have on the forces at the droplet surface. The droplet position and mean kinetic energy of the droplet are also investigated and compared to other works, proving the dynamics of a droplet motion found here.

hydrophobic environment triggering reactive fluorescence probe to real-time monitor mitochondrial DNA

摘要: Mitochondrial DNA has a special structure that is prone to damage resulting in many serious diseases, such as genetic diseases and cancers. Therefore, the rapid and specific monitoring of mitochondrial DNA damage is urgently needed for biological recognition. Herein, we constructed an in situ hydrophobic environment-triggering reactive fluorescence probe named MBI-CN. The fluorophore was 2-styrene-1H-benzo[d]imidazole, and malononitrile was introduced as a core into a molecule to initiate the hydrolysis reaction in the specific environment containing damaged mitochondrial DNA. In this design, MBI-CN conjugates to mitochondrial DNA without causing additional damages. Thus, MBI-CN can be hydrolyzed to generate MBI-CHO in an in situ hydrophobic environment with mitochondrial DNA damage. Meanwhile, MBI-CHO immediately emitted a significative fluorescence signal changes at 437 and 553 nm within 25 s for the damaged mitochondria DNA. Give that the specific and rapid response of MBI-CN does not cause additional damages to mitochondrial DNA, it is a potentially effective detection tool for the real-time monitoring of mitochondrial DNA damage during cell apoptosis and initial assessment of cell apoptosis.

Serum mitochondrial tsRNA serves as a novel biomarker for hepatocarcinoma diagnosis

摘要: Hepatocellular carcinoma (HCC), which makes up the majority of liver cancer, is induced by the infection of hepatitis B/C virus. Biomarkers are needed to facilitate the early detection of HCC, which is often diagnosed too late for effective therapy. The tRNA-derived small RNAs (tsRNAs) play vital roles in tumorigenesis and are stable in circulation. However, the diagnostic values and biological functions of circulating tsRNAs, especially for HCC, are still unknown. In this study, we first utilized RNA sequencing followed by quantitative reverse-transcription PCR to analyze tsRNA signatures in HCC serum. We identified tRF-Gln-TTG-006, which was remarkably upregulated in HCC serum (training cohort: 24 HCC patients vs. 24 healthy controls). In the validation stage, we found that tRF-Gln-TTG-006 signature could distinguish HCC cases from healthy subjects with high sensitivity (80.4%) and specificity (79.4%) even in the early stage (Stage I: sensitivity, 79.0%; specificity, 74.8%; 155 healthy controls vs. 153 HCC patients from two cohorts). Moreover, in vitro studies indicated that circulating tRF-Gln-TTG-006 was released from tumor cells, and its biological function was predicted by bioinformatics assay and validated by colony formation and apoptosis assays. In summary, our study demonstrated that serum tsRNA signature may serve as a novel biomarker of HCC.

NETO2 promotes melanoma progression via activation of the Ca/CaMKII signaling pathway

摘要: Melanoma is the most aggressive cutaneous tumor. Neuropilin and tolloid-like 2 (NETO2) is closely related to tumorigenesis. However, the functional significance of NETO2 in melanoma progression remains unclear. Herein, we found that NETO2 expression was augmented in melanoma clinical tissues and associated with poor prognosis in melanoma patients. Disrupting NETO2 expression markedly inhibited melanoma proliferation, malignant growth, migration, and invasion by downregulating the levels of calcium ions (Ca2+) and the expression of key genes involved in the calcium signaling pathway. By contrast, NETO2 overexpression had the opposite effects. Importantly, pharmacological inhibition of CaMKII/CREB activity with the CaMKII inhibitor KN93 suppressed NETO2-induced proliferation and melanoma metastasis. Overall, this study uncovered the crucial role of NETO2-mediated regulation in melanoma progression, indicating that targeting NETO2 may effectively improve melanoma treatment.

Conserved gene arrangement in the mitochondrial genomes of barklouse families Stenopsocidae and Psocidae

摘要: Substantial variation in gene organization and arrangement has been reported for sequenced mitochondrial (mt) genomes from the suborders of the insect order Psocoptera. In this study we sequenced the complete mt genome of , the first representative of the family Stenopsocidae from the suborder Psocomorpha. Relative to the ancestral pattern, rearrangements of a protein-coding gene ( ) and five tRNA genes ( , , , , ) were found. This pattern was similar to that of two barklice from the family Psocidae, with the exception of the translocation of , and Based on comparisons of pairwise breakpoint distances of gene rearrangements, gene number and chromosome number, it was concluded that mt genomes of Stenopsocidae and Psocidae share a relatively conserved pattern of gene rearrangements; mt genomes within the Psocomorpha have been generally stable over long evolutionary history; and mt gene rearrangement has been substantially faster in the booklice (suborder Troctomorpha) than in the barklice (suborders Trogiomorpha and Psocomorpha). It is speculated that the change of life history and persistence of unusual reproductive systems with maternal inheritance contributed to the contrasting rates in mt genome evolution between the barklice and booklice.

N-Positive ion activated rapid addition and mitochondrial targeting ratiometric fluorescent probes

摘要: Heterocyclic compound quinoline and its derivatives exist in natural compounds and have a broad spectrum of biological activity. They play an important role in the design of new structural entities for medical applications. Similarly, indoles and their derivatives are found widely in nature. Amino acids, alkaloids and auxin are all derivatives of indoles, as are dyes, and their condensation with aldehydes makes it easy to construct reaction sites for nucleophilic addition agents. In this work, we combine these two groups organically to construct a rapid response site (within 30 s) for H S, and at the same time, a ratiometric fluorescence response is presented throughout the process of H S detection. As such, the lower detection limit can reach 55.7 nmol/L for H S. In addition, cell imaging shows that this probe can be used for the mitochondrial targeted detection of endogenous and exogenous H S. Finally, this probe application was verified by imaging H S in nude mice.

Effect of K and Ca on catalytic activity of Mn-CeO

摘要: Mn-CeO /Ti-pillared clay (PILC) is an attractive catalyst for selective catalytic reduction of NO at low temperature because of its low cost. The poisoning of K and Ca on the catalyst of Mn-CeO /Ti-PILC is an important problem because K and Ca are always in presence in flue gas. To investigate the effect of K and Ca on the physicochemical characters of the catalysts, the techniques of NH -temperature programmed desorption (TPD), H -temperature programmed reduction (TPR), and X-ray photoelectron spectroscopy (XPS) were used to analyze the fresh and deactivated catalysts of Mn-CeO /Ti-PILC. (Ca)Mn-CeO /Ti-PILC and (K)Mn-CeO /Ti-PILC are denoted for the dopes of the catalyst of Mn-CeO /Ti-PILC with Ca and K, respectively. The activities of Mn-CeO /Ti-PILC, (Ca)Mn-CeO /Ti-PILC and (K)Mn-CeO /Ti-PILC for NH -selective catalytic reduction (SCR) reaction at low temperature were investigated. The results showed that with the dopes of K and Ca on the catalysts, the SCR activities of the catalysts decreased greatly, and K exhibited more poisoning effect than Ca. With the dopes of K and Ca, the acidity, the redox property and chemisorbed oxygen on the surfaces of the catalysts were decreased, which resulted in a decreasing in SCR activity.

Protective roles of heat stress on the neurons in hippocampal CA1 region of mice

摘要: The effects of heat stress on the neurons in hippocampal CA1 region of brain ischemia/reperfusion were explored. The mice were pretreated with heat stress followed by ischemia/reperfusion by clipping bilateral cervical commo n arteries for 7 min. Mice were divided randomly into four groups as follows: (1) normal control group; (2) heat stress pretreated subsequent to ischemia/reperfusion group (HS/IR); (3) ischemia/reperfusion group (IR); and (4) heat stress group (HS). Animals in the last three groups were subdivided into three subgroups: 1 d, 4 d, 14 d respectively. The Morris water maze was used to test the ability of learning and memorizing, Nissl staining was used to count the average number of survived neurons in hippocampal CA1 region, and immunohistochemistry combined with image analysis system to detect the changes of Microtubule associated protein 2 (MAP-2) expression. The results showed that mice in IR group exhibited increased escape latency when compared with that of normal, HS and HS/IR groups (<0.01), and the mice in IR group adopted an inefficient search strategy, major in circling and restricted searching manners. Nissl staining results showed a significant reduction in the number of pyramidal neurons in hippocampal CA1 regions in HS/IR and IR groups, with a decrease in IR group (<0.01). Compared with normal group, the expression of MAP-2 in hippocampal CA1 region obviously decreased in IR group (<0.05). The present results indicate that heat stress pretreatment can improve the spatial learning and memorizing function through protection to hippocampal neurons.

聚变-裂变能源混合堆可行性及在我国核能发展中作用的分析

简要论述了核能在我国能源发展战略中的地位及聚变-裂变混合堆在核能持续发展中的重要作用。对以不久将来即可实现的ITER聚变装置作驱动堆芯、天然铀水冷裂变系统作包层的混合堆做了细致的分析。这种混合堆型可以实现GWe级净电功率输出,年造钚1 656 kg,支持2.68个同功率压水堆电站对易裂变燃料的需要。初步的经济评估说明,混合堆电的成本是同功率压水堆电成本的1.67倍;而在不计燃料成本的情况下,混合堆与压水堆组合系统电的成本是同功率压水堆电成本的1.18倍。考虑到一般压水堆需消耗大量的天然铀,加上铀浓缩成本,混合堆与压水堆组合系统电的成本,与压水堆电的成本是可以相比拟的。

Laboratory study on high-temperature adsorption of HCl by dry-injection of Ca(OH)

摘要: Combustion-generated hydrogen chloride (HCl) is considered to be a very hazardous acid gaseous pollutant. This paper presents a laboratory study on the dry adsorption of HCl. The experiments were conducted in a dual-layer granular bed filter, at gas temperatures of 500°C–700°C and (Ca)/ (Cl)molar ratios of 1.0–5.0 using the silver nitrate titration method by dry adsorbent powders Ca(OH) . Mainly, the adsorption efficiency of HCl and utilization efficiency of Calcium were studied, by varying relevant factors including (Ca)/ (Cl), temperature, feeding method, water vapor and CO . With a relatively higher HCl concentration of 1000 ppm, the experimental results revealed that 600°C may be the optimum temperature for HCl adsorption when optimum (Ca)/ (Cl) was 2.5 in our tests. The results also demonstrated that the feeding at a constant pressure was more effective, and the HCl adsorption efficiency could rapidly reach over 90% with (Ca)/ (Cl) = 2.5 at 600°C. Furthermore, the HCl adsorption efficiency was found to be slightly promoted by water vapor, while could be impeded by CO , and the utilization efficiency of calcium could be up to 74.4% without CO , while was only 36.8% with CO when (Ca)/ (Cl) was 2.5 at 600°C.

摘要: 本文提出了一种快速响应客户需求的产品优化设计方法,通过基于模糊相似理论的需求信息模糊聚类方法,对具有模糊性和多样性的原始客户需求信息进行精简分类处理,以获取客户需求集合;然后构建了一种综合联合分析法(CA)和质量功能展开(QFD)的全新CA-QFD需求转化方法,从客户需求到产品设计属性进行精确转化,并通过对实例产品的优化设计,实现快速响应客户需求;最后,通过锻压机主工作液压缸的优化设计实例,验证了本文方法的可行性和适用性

NRTIs’ effect on the sequence of mitochondrial DNA HV 2 in HIV infected patients

摘要: Potent combination antiretroviral therapy (cART) has significantly improved the life expectancy of people living with human immunodeficiency virus (HIV), but it has many side effects such as lipodystrophy (LD), hepatic steatosis, and lactic acidosis. Nucleoside reverse transcriptase inhibitors (NRTIs) could damage the mitochondria by inhibiting the human DNA polymerase gamma, leading to mtDNA deletion. However, it remains uncertain whether NRTIs could induce the hypervariable region (HV) mutations of the D loop of mitochondria in Chinese HIV/AIDS patients, and whether that effect is different between individuals with and without LD. Hereby, 30 Chinese AIDS patients who were receiving antiretroviral drugs were recruited, among which 16 had symptomatic LD and 14 did not. Blood samples were collected prior to and after 96 weeks of treatment. Total DNA was extracted from peripheral blood mononuclear cells (PBMCs). Fragments of 728 bp in length containing HV 2 were amplified by standard polymerase chain reaction (PCR). Direct DNA-sequencing analysis techniques were used to detect mitochondrial sequence variants between paired longitudinal samples. Alterations were compared with the revised Cambridge Reference Sequence (rCRS) to determine mutation or polymorphism. Results showed that two years after ART, totally seven cases exhibited sequence variations, five individuals showed 73€A→G revised variation (two with and three without LD), while two cases of LD were found to have other nucleotide alterations. There was no new alteration in individuals without LD. In conclusion, NRTIs could induce mutation of mtDNA HV 2, which might contribute to the development of LD.
